Stimulated Raman scattering microscopy with spectral phasor analysis: applications in assessing drug-cell interactions William Tipping, The University of Strathclyde This data was generated for the publication of a research article of the same title and investigates the phenotypic effects of statin treatment in breast cancer cells using stimulated Raman scattering microscopy and spectral phasor analysis. Raman spectral data was acquired using a Renishaw InVia microscope system controlled using Renishaw WiRE 4.4 software. Spectral and imaging datasets are collated as Excel file format (.exe). The data has been post-processed in WiRE 4.4 and plotted on Origin2018, the files are collated in .exe format with a different tab per cell line used. SRS images were acquired using a Leica multiphoton microscope. SRS images are provided in .tif file format with post-processing performed using ImageJ or .lif format that is compatible with ImageJ.. Figure 1 The data is provided in .tif format and the plot of mean 2851 cm-1 intensity per cell is collated in .exe format. Figure 2 The images are provided in .tif file format with scale bars: 10 um. The spectral phasor plot was performed using a feely available plugin for imageJ. Figure 3 For each cell line, the Average intensity projection, spectral phasor plot, lipid droplet and lipid droplet periphery images are provided in .tif format. The normalised spectra are provided in .exe format. Figure 4 The percentage area and ratio 3015/2965 cm-1 data are provided in separate .exe files.
