Temporal imaging of drug dynamics in live cells using stimulated Raman scattering microscopy and a perfusion cell culture system William Tipping, The University of Strathclyde This data was generated for the publication of a research article of the same title and investigates the intracellular detection of the discoidin domain receptor 1 inhibitor, 7RH in a panel of cancer cell lines using stimulated Raman scattering microscopy. Raman spectral data was acquired using a Renishaw InVia microscope system controlled using Renishaw WiRE 4.4 software. Spectral and imaging datasets are collated as Excel file format (.exe). The data has been post-processed in WiRE 4.4 and plotted on Origin2018, the files are collated in .exe format. SRS images were acquired using a Leica SP8 multiphoton microscope. SRS images are provided in .tif file format with post-processing performed using ImageJ or in .lif format that is compatible with ImageJ. Figure 1 The raw Raman spectral data is provided in .wdf format compatible with WiRE 4.4 and an .exe file collates the raw and baseline subtracted data. The ratiometric images were produced using Matlab and are provided in .ppt format. The Raw mapping data is provided in .wdf and .spc format that is compatible with Matlab.
